目的观察槐耳颗粒对单侧输尿管梗阻模型(UUO)肾组织beclin-1、FN的影响来进一步阐明槐耳在急性肾损伤中作用的可能机制。方法 Wistar大鼠随机分为假手术组(Shan组),模型组(UUO组),模型组+槐耳治疗组(UHE组),造模后0、3、7天分析大鼠肾组织病理损伤,血生化Scr、Urea值,beclin-1、FN的表达分布及变化。结果 1造模后7天,UUO组较Sham组病理损伤评分和血生化指标Scr、Urea均有显著上调(P<0.05)。2造模后3天、7天,UUO组beclin-1、FN在肾小管上皮细胞中表达上调(P<0.05)。3造模后3天、7天,槐耳治疗组较模型组Scr、Urea、beclin-1、FN均有显著下调(P<0.05)。结论梗阻性肾损伤中,槐耳可以抑制肾组织中自噬的发生,从而减缓小管间质纤维化的形成,延缓小管间质的损伤进程。
Objective To observe the effect of Huaier granules on the expression of beclin-1 and FN in kidney of unilateral ureteral obstruction (UUO) model to further elucidate the possible mechanism of Huaier in acute renal injury. Methods Wistar rats were randomly divided into Sham group, model group (UUO group) and model group + Huaier treatment group (UHE group). The rats were sacrificed on day 0, , Blood biochemical Scr, Urea value, beclin-1, FN expression and distribution changes. Results 1, 7 days after modeling, pathological injury score and blood biochemical markers Scr and Urea of UUO group were significantly higher than Sham group (P <0.05). The expressions of beclin-1 and FN in UUO group were upregulated in renal tubular epithelial cells 3 days and 7 days after model establishment (P <0.05). 3, 3 days and 7 days after modeling, Scr, Urea, beclin-1 and FN were significantly decreased in the Huaier group compared with the model group (P <0.05). Conclusions In obstructive kidney injury, Huai Er can inhibit the occurrence of autophagy in renal tissue, thereby reducing the formation of tubulointerstitial fibrosis and delaying the process of tubulointerstitial injury.
