目的 观察大川芎方对偏头痛大鼠模型疼痛行为学及SP等物质的影响,评价和探讨大川芎方治疗偏头痛的作用机制.方法 各组大鼠不同药物连续灌胃3d,最后一次灌胃后1h造模,记录大鼠的疼痛行为学表现并检测其颈静脉血中CGRP、SP、VIP物质的浓度.结果 大川芎方各组大鼠的疼痛行为学表现较生理盐水组减少,高剂量组更为显著(P<0.05);各组间血浆SP、CGRP浓度无统计学差异;大川芎方各组能升高偏头痛模型大鼠血浆NPY水平,高剂量组更为显著(P<0.05).结论 大川芎方可能是通过升高血浆中NPY的浓度发挥镇痛作用来治疗偏头痛的.“,”Objective Investigate the influence of Dachuanxiong Fang on nociceptive behaviors,substance P and other vasoactive substances in external jugular vein in rats after electrical stimulation of dura mater surrounding the superior sagittal sinus,evaluate and explore the mechanism of Dachuanxiong Fang in the treatment of migraine.Methods Rats in each group were given intragastric administration of different drugs for 3 days,and the rats were sacrificed at 1 hour after the last administration.Nociceptive behaviors were observed and the concentration of CGRP,SP,NPY were detected by enzyme linked immunosorbent assay.Results The nociceptive behaviors of the rats in the Dachuanxiong Fang group was less than that in the saline group while the high-dose groups were significantly more (P<0.05);There were no statistical differences in SP and CGRP levels between each group.The NPY levels were inordinately increased in groups treated with Dachuanxiong Fang compared with the saline group.High-dose group was significantly more (P<0.05).Conclusion The effect of Dachuanxiong Fang in migraine treatment may due to the up regulation of NPY in plasma.
