目的 研究一氧化氮合成酶 (nitricoxidesynthase ,NOS)的三种同工酶在大鼠坐骨神经中的表达和分布情况。方法 取 10只SD大鼠双侧正常坐骨神经 ,一侧用于Western印迹检查 ,另外一侧用于免疫组化检查。结果 Western印迹法中 ,用神经元型一氧化氮合成酶 (neuronalNOS ,nNOS)单抗和内皮细胞型一氧化氮合成酶 (endothelialNOS ,eNOS)单抗 ,分别发现一个相对分子质量为 15 5 0 0 0和一个相对分子质量为 14 0 0 0 0的蛋白带 ,分别与nNOS和eNOS的阳性对照相一致。免疫组化显示 ,nNOS存在于大多数Schwann细胞和轴突中 ,eNOS存在于坐骨神经内部的血管内皮细胞。结论 正常大鼠的坐骨神经中有NOS ,其中nNOS存在于Schwann细胞和轴突中 ,eNOS仅存在于血管内皮细胞。
Objective To study the expression and distribution of three isoenzymes of nitric oxide synthase (NOS) in rat sciatic nerve. Methods Bilateral normal sciatic nerve of 10 SD rats were obtained. One side was used for Western blot examination and the other side was used for immunohistochemistry. Results Western blotting showed that a monoclonal antibody against neuronal NOS (nNOS) and endothelial nitric oxide synthase (eNOS) was found to have a relative molecular weight of 15 500 0 and a protein band with a molecular weight of 140,000, consistent with the positive control of nNOS and eNOS, respectively. Immunohistochemistry showed that nNOS was present in most Schwann cells and axons and eNOS was present in vascular endothelial cells inside the sciatic nerve. Conclusion There are NOS in the sciatic nerve of normal rats. Among them, nNOS is found in Schwann cells and axons, while eNOS is only found in vascular endothelial cells.
