为探讨含有透明质酸钠和白蛋白的卡铂复合液对腹腔内化疗后腹腔粘连的抑制作用,采用斑点杂交和原位杂交方法观察其对大鼠腹腔内化疗后腹膜组织Ⅰ型和Ⅲ型胶原mRNA水平的影响。结果显示卡铂复合液腹腔内化疗后第21天和第48天大鼠腹膜α1(Ⅰ)和α1(Ⅲ)胶原mRNA水平明显低于卡铂液组(P<0.01)。腹腔粘连发生率和粘连程度显著低于卡铂液组(P<0.01)。卡铂液组大鼠腹腔内化疗后腹膜明显增厚,间皮下成纤维细胞增生活跃,细胞内可见大量的银颗粒;而卡铂复合液组大鼠腹膜下成纤维细胞增生明显受到抑制,银颗粒数量明显减少。研究表明卡铂复合液腹腔内化疗对胶原基因表达的增加有明显抑制作用,可明显地减少成纤维细胞胶原合成和抑制腹腔粘连形成。
To investigate the inhibitory effect of carboplatin complex containing sodium hyaluronate and albumin on intraperitoneal adhesions after intraperitoneal chemotherapy, spot hybridization and in situ hybridization were used to observe the type I and type III peritoneal tissues after intraperitoneal chemotherapy in rats. The effect of collagen mRNA levels. The results showed that the mRNA levels of α1(1) and α1(III) collagen in the peritoneum of carboplatin-treated rats on the 21st and 48th days after intraperitoneal chemotherapy were significantly lower than those in the carboplatin group (P<0.01). The incidence and degree of adhesion in the abdominal cavity were significantly lower than those in the carboplatin group (P<0.01). In the carboplatin group, the peritoneum was significantly thickened after intraperitoneal chemotherapy, and the proliferation of submucous fibroblasts was active. A large number of silver particles were seen in the cells. The proliferation of subperitoneal fibroblasts in the carboplatin group was significantly inhibited. The number of particles is significantly reduced. Studies have shown that intraperitoneal chemotherapy with carboplatin has significantly inhibited the increase of collagen gene expression, and can significantly reduce fibroblast collagen synthesis and inhibit the formation of intraperitoneal adhesions.
