目的:通过对露水草愈伤组织诱导与分化条件的研究,为露水草快速繁殖及转基因研究奠定基础。方法:利用正交设计方法对影响露水草愈伤组织诱导与分化的因素进行优化。结果:1.影响露水草愈伤组织诱导率的主要因素是生长素种类和外植体部位,生长素用NAA或2,4-D优于IAA和IBA,外植体用叶鞘部位极显著优于种子、根和叶;细胞分裂素种类和活性炭(AC)浓度对愈伤诱导率影响不显著。2.影响愈伤组织分化成芽的主要因素是愈伤组织的来源,根和叶形成的愈伤组织成芽率显著优于种子和叶鞘形成的愈伤组织;生长素、细胞分裂素种类和AC浓度对成芽率影响不显著。3.影响外植体直接成苗的主要因素是外植体部位,种子作外植体极显著优于叶鞘、叶和根;生长素、细胞分裂素种类和AC浓度对成苗率影响不显著。4.影响不定根诱导形成的主要因素是生长素种类,生长素用IBA成根率极显著优于IAA和NAA;6-BA和AC浓度对成根率影响不显著。结论:通过该项研究,露水草愈伤组织诱导与分化的条件得到较好优化。
OBJECTIVE: To study the conditions of callus induction and differentiation of dew grass and lay the foundation for the rapid propagation and transgenic studies of dew grass. Methods: The orthogonal design method was used to optimize the factors affecting the callus induction and differentiation of dew grass. The main factors affecting the callus induction rate of P. delavayi were auxin species and explants. NAA or 2,4-D auxin was superior to IAA and IBA. In the seeds, roots and leaves; cytokinin and activated carbon (AC) concentration on callus induction rate was not significant. 2. The main factor influencing the differentiation of callus into bud is the origin of callus, the callus formation rate of callus formed by root and leaf is significantly better than the callus formed by seed and leaf sheath; the content of auxin, cytokinin and AC concentration had no significant effect on the percentage of budding. The main factors that affect the direct seedling of explants are explants, the seed explants are significantly better than the leaf sheath, leaf and root; auxin, cytokinin and AC concentration had no significant effect on the percentage of seedlings . The main factors affecting the induction of adventitious root formation are auxin types. The rooting rate of auxin with IBA is significantly superior to that of IAA and NAA. The concentration of 6-BA and AC has no significant effect on the percentage of rooting. Conclusion: Through the study, the conditions of callus induction and differentiation of Dew grass are well optimized.
