目的研究卡巴胆碱对肠缺血再灌流动物脏器功能的保护作用及对肠组织和白细胞炎症介质表达的影响。方法检测肠缺血再灌流大鼠和肠部分缺血再灌流兔伤后脏器功能指标、白细胞中细胞因子表达和血浆、肠组织TNFα含量。结果卡巴胆碱治疗组大鼠再灌流2h后血浆ALT、BUN和CK低于对照组,白细胞中TNFα表达下调,IFN、IL4和IL10表达上调;治疗组兔血浆ALT、Cr和CKMB在缺血和再灌流早期增加幅度低于实验组,再灌流后趋于恢复,实验组渐升高;肠组织TNFα含量低于实验组。结论卡巴胆碱改善肠缺血再灌流动物脏器功能,抑制肠组织TNFα的合成,调节白细胞致炎和抗炎因子的表达比例。
Objective To investigate the protective effect of carbachol on visceral ischemia-reperfusion injury and its effect on intestinal tissue and leukocyte inflammatory mediators. Methods The indexes of organ function after ischemia and reperfusion in rabbits with intestinal ischemia and reperfusion and the expression of cytokines in leukocytes and TNFα in plasma and intestine were detected. Results The levels of ALT, BUN and CK in plasma of rats in carbachol treatment group were lower than that in control group 2h after reperfusion, the expression of TNFα was down-regulated and the levels of IFN, IL-4 and IL10 were up-regulated in leukocytes; In the early stage of reperfusion, the increase rate was lower than that in the experimental group, then recovered after reperfusion, and the experimental group was gradually increased. The content of TNFα in the intestinal tissue was lower than that in the experimental group. Conclusions Carbachol can improve the organ function of intestinal ischemia-reperfusion animals, inhibit the synthesis of TNFα in intestinal tissue and regulate the ratio of inflammatory and anti-inflammatory cytokines.
