目的 探讨洛沙坦对大鼠硝酸甘油 (NTG)耐药的改善作用。方法 雄性Wistar大鼠 2 8只 ,随机分成 4组。对照组 (C组 ,n =10 ) ,硝酸甘油组 (NTG组 ,n =6 ) ,洛沙坦组 (Losartan组 ,n =6 ) ,硝酸甘油 +洛沙坦组 (NTG +Losartan组 ,n =6 )。通过大鼠离体主动脉环对不同浓度硝普钠 (SNP)的舒张反应及离体主动脉环舒张 5 0 %所需SNP的浓度 (EC50 ) ,观察各组大鼠对SPN的降压反应。用放射免疫方法测定血浆和血管组织的内皮素 1(ET 1)的含量。结果 NEG +Lossartan组大鼠对SNP的降压反应明显强于NTG组大鼠 ,NTG +Losartan组大鼠离体主动脉环的舒张反应较NTG组大鼠有明显的改善 ,EC50 降低。NTG +Losartan组大鼠血浆及血管组织ET 1含量均较NTG组大鼠明显下降。结论 血管紧张素Ⅱ受体拮抗剂Losartan可以改善NTG的耐药。其耐药的改善可能与ET 1降低有关
Objective To investigate the effects of losartan on the drug resistance of rat NTG. Methods Male Wistar rats, 28 were randomly divided into 4 groups. The rats in control group (group C, n = 10), nitroglycerin group (NTG group, n = 6), losartan group (n = 6), nitroglycerin + losartan group = 6). The relaxation responses of isolated aorta rings to different concentrations of SNP and the required concentration of SNP for 50% of isolated aortic rings were observed. The antihypertensive response to SPN in each group was observed . The contents of endothelin 1 (ET 1) in plasma and vascular tissue were determined by radioimmunoassay. Results The antihypertensive response to SNP in rats in NEG + Lossartan group was significantly stronger than that in NTG rats. The relaxation of isolated aortic rings in rats in NTG + Losartan group was significantly improved compared with that in NTG rats and EC50 was decreased. NTG + Losartan group plasma and vascular ET 1 levels were significantly lower than the NTG group rats. Conclusion Losartan, an angiotensin Ⅱ receptor antagonist, can improve the drug resistance of NTG. The drug resistance may be related to the reduction of ET 1
