目的探讨银杏叶提取物(EGb)的抗自由基保护心血管功能及其舒张离体脑血管的作用与释放NO的关系。方法(1)家兔心脏与离体主动脉环用二苯三硝基苯肼(DPPH)自由基造成心收缩功能障碍和乙酰胆碱诱发的血管舒张功能损伤与经EGb预处理后再用DPPH损伤组比较。(2)观察EGb对猪离体基底动脉环的舒张作用及其对电刺激诱发的舒张作用的影响。结果(1)EGb能保护心脏与血管抗DPPH自由基对心收缩功能与血管内皮细胞舒张功能的损伤;(2)EGb对猪基底动脉有量效依赖性的舒张作用,舒张内皮细胞健全的基底动脉的作用比除去内皮细胞的基底动脉更显著。(3)EGb能增强电刺激基底动脉诱发的舒张作用,此作用可被N硝基精氨酸或河豚素毒所阻断。结论EGb能保护家兔离体心脏与血管内皮细胞抗DPPH自由基损伤。对离体基体动脉有舒张作用,并能增强电刺激诱发的舒张作用,此作用与其促进NO释放有关。
Objective To investigate the anti-free radical protective effects of Ginkgo biloba extract (EGb) on cardiovascular function and the effect of relaxing diastolic cerebral blood vessels on NO release. Methods (1) The cardiac contractile dysfunction induced by DPPH free radical and the vasodilation function induced by acetylcholine in rabbit heart and isolated aortic rings were pretreated with EGb and then with DPPH injury group Compare (2) The effect of EGb on the relaxation of porcine isolated basilar artery rings and its effect on the relaxation induced by electrical stimulation were observed. Results (1) EGb could protect cardiac and vascular anti-DPPH free radical contractile function and vascular endothelial cell diastolic function. (2) EGb had a dose-dependent relaxation effect on porcine basilar artery and relaxation of endothelial cells The effect of the artery is more pronounced than that of the basilar artery, which removes endothelial cells. (3) EGb can enhance the vasodilatation induced by electrical stimulation of the basilar artery, and this effect can be blocked by N-nitroarginine or guinea pig venom. Conclusion EGb can protect rabbit heart and vascular endothelial cells against DPPH free radical injury. On the isolated basilar artery has a relaxation effect, and can enhance the electrical stimulation-induced relaxation, this role and its promotion of NO release.
