目的:探讨补阳还五汤防治经皮腔内冠脉成形术(PTCA)后再狭窄的消长机制。方法:仿PTCA术后再狭窄的新西兰兔模型28只,以原位引物标记法测动脉内皮钢-锌超氧化物歧化酶(SOD-1)及血小板行化生长因子受体(PDGFR)的基因表达,分析再狭窄消长机理。结果:主动脉球囊扩张术后7天,手术部位动脉内皮的SOD-1基因表达增强,与对照组有显著差异(P<0.05),而PDGFRmRNA的表达则减弱,与对照组也有显著性差异(P<0.05)。结论:补阳还五汤增强动脉内皮SOD-1的基因表达,改善缺血再灌注的供氧。同样情况下,补阳还五汤抑制PDGFRmRNA表达,以控制细胞的病理性增殖。
Objective: To investigate the mechanism of Buyang Huanwu Decoction in preventing and treating the restenosis after percutaneous transluminal coronary angioplasty (PTCA). METHODS: Twenty-eight New Zealand rabbit models of post-PTCA restenosis were used to measure the genes of arterial endothelial steel-zinc superoxide dismutase (SOD-1) and platelet growth factor receptor (PDGFR) by in situ primer labeling. Express and analyze the mechanism of restenosis. RESULTS: At 7 days after aortic balloon dilatation, the SOD-1 gene expression in the endothelium of the surgical site was increased, which was significantly different from that in the control group (P<0.05). The expression of PDGFR mRNA was attenuated, which was also significant in the control group. Sexual difference (P<0.05). Conclusion: Buyang Huanwu Decoction enhances the expression of SOD-1 gene in endothelial and improves the oxygen supply during ischemia-reperfusion. Under the same circumstances, Buyang Huanwu Decoction inhibits PDGFR mRNA expression to control the pathological proliferation of cells.
