目的研究注射用复方葛根素粉针中葛根素在Beagle犬体内药动学。方法建立快速敏感的高效液相色谱检测方法测定Beagle犬血中葛根素,以6只Beagle犬交叉注射给药,药-时曲线数据经3P97药动学计算程序处理。结果葛根素的药-时过程符合二室模型,葛根素注射液犬体内葛根素的t1/2α=3.111±2.166 h,t1/2β=54.946±32.475 h,AUC0-t=2228.292±1363.136 mg.h-1.L-1;注射用复方葛根素粉针犬体内葛根素的t1/2α=3.752±0.572 h,t1/2β=56.247±3.112 h,AUC0-t=2361.020±109.238 mg.h-1.L-1,结果表明,两者药动学过程没有统计学差异。结论注射用复方葛根素粉针中葛根素与牛磺酸配伍对葛根素的体内药动学无影响。
Objective To study the pharmacokinetics of puerarin in injection puerarin injection in Beagle dogs. Methods Fast and sensitive high performance liquid chromatography (HPLC) was used to determine the puerarin in blood of Beagle dogs. Six Beagle dogs were injected intraperitoneally. The drug-time curve data were processed by 3P97 pharmacokinetic calculation. Results The puerarin-time course was in accordance with the two-compartment model. The puerarin in puerarin injection was 3.111 ± 2.166 h, t1 / 2β = 54.946 ± 32.475 h, AUC0-t = 2228.292 ± 1363.136 mg.h -1.L-1. The puerarin in the compound puerarin injection for dogs showed t1 / 2α = 3.752 ± 0.572 h, t1 / 2β = 56.247 ± 3.112 h, AUC0-t = 2361.020 ± 109.238 mg · h-1. L-1, the results show that there is no statistical difference between the two pharmacokinetic process. Conclusion The combination of puerarin with taurine in puerarin injection powder injection has no effect on the pharmacokinetics of puerarin in vivo.
