目的 :探讨基质金属蛋白酶 - 2 (MMP- 2 )、组织转化生长因子 β1 (TGFβ1 )的表达以及微血管密度 (MVD)与大肠癌临床生物学特性的相关性。方法 :应用标准霉菌抗生物素蛋白 -过氧化物酶亲和免疫组织化学法 ,检测 6 0例大肠癌标本中MMP- 2、TGFβ1的表达及 MVD值。结果 :MMP- 2阳性表达率 5 6 .7% (34/6 0 ) ;TGFβ1的阳性表达率 4 3.3% (2 6 /6 0 ) ;MVD1 8~ 1 32 ,平均值 4 6 .6± 2 0 .8。无淋巴结转移组、高分化组、无远处转移组的 MMP- 2和 TGFβ1表达水平分别低于有淋巴结转移组、低分化组、有远处转移组 ,(P <0 .0 5 )。 MVD值增大提示大肠癌分化程度低、易发生淋巴结转移和远处转移及预后差 ;TGFβ1表达与大肠癌血管生成有关 ;MMP- 2、TGFβ1高表达组 MVD值大于 MMP- 2、TGFβ1低表达组 (P <0 .0 5 )。结论 :MMP- 2、TGFβ1在大肠癌的发生、发展过程中起重要作用 ,尤其在肿瘤侵袭、转移过程中发挥重要作用 ,MMP- 2、TGFβ1与 MVD关系密切
OBJECTIVE: To investigate the relationship between the expression of matrix metalloproteinase 2 (MMP-2), transforming growth factor-β1 (TGFβ1) and the microvessel density (MVD) and the clinical biological characteristics of colorectal cancer. Methods: The expression of MMP-2, TGFβ1 and MVD in 60 specimens of colorectal cancer were detected by standard mold avidin-peroxidase immunohistochemistry. Results: The positive expression rate of MMP-2 was 56.7% (34/60), the positive expression rate of TGFβ1 was 43.3% (26/60), MVD1 8-132 (average 46.6 ± 2) 0 .8. The expression of MMP-2 and TGFβ1 in non-lymph node metastasis group, well-differentiated group and distant metastasis group were lower than that in lymph node metastasis group, poorly differentiated group and distant metastasis group (P <0.05). The increased MVD value suggests that the differentiation degree of colorectal cancer is low, lymph node metastasis and distant metastasis are prone to occur, and the prognosis is poor. The expression of TGFβ1 is related to the angiogenesis of colorectal cancer. The MVD of MMP-2 and TGFβ1 overexpression group is higher than that of MMP-2 and TGFβ1 Group (P <0.05). Conclusion: MMP-2 and TGFβ1 play an important role in the carcinogenesis and progression of colorectal cancer, especially in the process of tumor invasion and metastasis.
