为减损排斥反应性淋巴细胞克隆,采用异型双功能试剂SPDP、2IT连接antiCD4、CD8单克隆抗体与天花粉毒素(TCS),应用SephacrylS200分离游离TCS,采用SDSPAGE电泳及细胞毒性分析测定合成的免疫毒素的纯度和生物学活性。电泳结果显示,连接混合物中含有连接的免疫毒素、游离的单克隆抗体和TCS,SephacrylS200可除去游离的TCS。体外,免疫毒素可特异性杀伤大鼠脾细胞,其作用具有剂量依赖性。由此提示抗CD4、CD8免疫毒素可以诱导免疫无反应性
