目的从分子水平揭示通莲I号方对食管癌细胞的抑制作用及其治疗噎膈的机制。方法体外培养食管鳞癌Eca109细胞,通莲I号方及3拆方分别作用于细胞,以MTT法检测细胞半数有效量,Western blot法观察NF-κB信号通路各基因表达变化情况。结果通莲I号方及活血行气、清热解毒拆方可显著抑制Eca109细胞增殖,通莲I号全方IC50=412 mg·L-1,清热解毒拆方IC50=718 mg·L-1,活血行气拆方IC50=693 mg·L-1;可抑制NF-κB信号通路相关分子IKKβ、NF-κB p65、TNF-α、IL-1β表达,作用强弱依次为:通莲I号全方>活血行气拆方>清热解毒拆方>滋阴养血拆方。结论通莲I号方用于治疗食管癌的有效组分在活血行气、清热解毒、滋阴养血类药物的协同作用,通过调节NF-κB介导的信号通路相关基因表达,抑制癌细胞增殖。
Objective To reveal the inhibitory effect of Tonglian I Prescription on esophageal cancer cells at the molecular level and its mechanism of treatment of the diaphragm. Methods Eca109 cells, Tonglian I and 3 cells were cultured in vitro. MTT assay was used to detect the half effective amount of cells. Western blot was used to observe the changes of NF-κB signaling pathway. The results Tonglian I Prescription and promoting blood circulation qi, Qingrejiedu Demobilization can significantly inhibit the proliferation of Eca109 cells, Tonglian I full of IC50 = 412 mg · L-1, detoxification IC50 = 718 mg · L-1, The expression of IKKβ, NF-κB p65, TNF-α and IL-1β in NF-κB signaling pathway was inhibited by IC50 = 693 mg · L-1, Prescription> Huoxue Qi gas demolition> detoxification Demolition> Ziyin nourishing demolition side. Conclusion Tonglian I-square is used to treat the synergistic effect of the active components of esophageal cancer on promoting blood circulation, clearing heat, detoxifying and nourishing yin and nourishing blood. By regulating NF-κB-mediated signaling pathway-related gene expression and inhibiting cancer cells proliferation.
