由于各组织细胞内均维持高GSH/GSSG比值状态,而离体样品中GSH又极易氧化为GSSG,因此测定GSSG时必须排除GSH干扰。通常采用巯基试剂作GSH隐蔽剂。本文比较了若干巯基试剂,并介绍了两种较适用的测定方法。方法一是适用于无蛋白液样品的乙二醛酶I(GL-I)法,GSSG回收率为94.0±11.2%;方法二是适用于血浆和无蛋白液样品的NEM过柱法,GSSG回收率为83.0±21.3%。
As cells in each tissue maintain a high GSH / GSSG ratio state, and in vitro samples of GSH can be easily oxidized to GSSG, GSH determination of GSH must be ruled out interference. Mercapto reagents are commonly used as GSH camouflage agents. This article compares several thiol reagents and introduces two more suitable assays. The first method is the glyoxalase I (GL-I) method which is suitable for protein-free samples and the recovery rate of GSSG is 94.0 ± 11.2%. The second method is the NEM column method and the GSSG recovery method for plasma and protein- The rate was 83.0 ± 21.3%.
