目的 :为了解糖尿病早期肾小管和集合管上皮细胞凋亡过程中ICE、Bcl 2、Bax、p5 3是否参与调控。方法 :雄性Wistar大鼠 6 0只 ,四氧嘧啶 (alloxan)尾静脉注射 (5 0mg/kg)造糖尿病模型 ;石蜡包埋肾切片经ICE、Bcl 2、Bax、p5 3免疫组化染色。结果 :ICE、Bcl 2、Bax、p5 3蛋白分别在肾小管和集合管上皮细胞两次凋亡发生前或发生时表达。结论 :p5 3蛋白的表达可能反应了高血糖致使肾小管、集合管上皮细胞DNA损伤 ;Bax表达增强可能有使Bcl 2与Bax的比值发生变化 ,以及在两次凋亡发生前ICE表达增强均与受损的肾小管、集合管上皮细胞凋亡有关。
OBJECTIVE: To investigate whether ICE, Bcl 2, Bax and p5 3 are involved in the regulation of the apoptosis of tubular and collecting duct epithelial cells in the early stage of diabetes mellitus. Methods: Sixty male Wistar rats were injected into the tail vein of alloxan (50 mg / kg). The paraffin-embedded sections were stained with ICE, Bcl 2, Bax and p5 3 immunohistochemistry. Results: The expressions of ICE, Bcl 2, Bax and p5 3 protein were detected before and at the occurrence of two apoptosis of renal tubular and collecting duct epithelial cells respectively. CONCLUSION: The expression of p5 3 protein may reflect the DNA damage of renal tubular and collecting duct epithelial cells induced by hyperglycemia. The increase of Bax may change the ratio of Bcl 2 to Bax, and the increase of ICE both before and after apoptosis And damaged tubules, collecting duct epithelial cell apoptosis related.
