【目的】观察具有益气解毒活络作用的中药复方对早期糖尿病肾病(DN)大鼠血清中葡葡糖转运蛋白1(GLUT1)含量的影响。【方法】采用高脂高糖加小剂量链脲佐菌素(STZ)的方法复制DN大鼠模型,将造模成功的Wistar大鼠按血糖值随机分为5组,即中药预防组,中药高、低剂量组,西药对照组,模型组,并设立正常组进行对照。实验结束后检测各组大鼠空腹血糖(FBG)、糖化血红蛋白(HbA1C)及24 h尿微量白蛋白(U-mAlb)含量。采用酶联免疫吸附反应(ELISA)法检测大鼠血清中GLUT1的含量,并取肾组织进行病理检查。【结果】与正常组比较,模型组的FBG、HbA1C、U-mAlb、血清GLUT1含量均显著升高(P<0.01);与模型组比较,各治疗组及预防组的各指标均显著降低(P<0.05或P<0.01);在改善血糖及U-mAlb方面,中药预防组效果最好,中药高剂量组次之(降血糖),西药对照组次之(降低U-mAlb);模型组大鼠血清中GLUT1含量最高,经治疗后,其含量显著下降,其中以中药预防组和中药低剂量组效果最好,西药对照组次之;各给药组大鼠肾组织病理变化程度明显轻于模型组。【结论】益气解毒活络中药复方防治早期糖尿病肾病的作用可能与其降低GLUT1在血清中的含量有关。
【Objective】 To observe the effect of traditional Chinese medicine (TCM) recipe, which has the function of benefiting qi toxin and detoxifying on the content of glucose transporter 1 (GLUT1) in the serum of early diabetic nephropathy (DN) rats. 【Methods】 DN rats were induced by high-fat, high-sugar and low-dose streptozotocin (STZ). Wistar rats were randomly divided into 5 groups according to their blood glucose levels, namely traditional Chinese medicine prophylaxis group, traditional Chinese medicine High and low dose group, Western medicine control group, model group, and established a normal control group. After the experiment, fasting blood glucose (FBG), glycosylated hemoglobin (HbA1C) and 24-hour urine microalbumin (U-mAlb) were detected. The content of GLUT1 in rat serum was detected by enzyme-linked immunosorbent assay (ELISA), and the renal tissue was taken for pathological examination. 【Results】 The levels of FBG, HbA1C, U-mAlb and GLUT1 in the model group were significantly increased compared with those in the normal group (P <0.01). Compared with the model group, the indexes of each treatment group and the prevention group were significantly decreased P <0.05 or P <0.01). The Chinese traditional medicine prophylaxis group had the best effect on improving blood glucose and U-mAlb, followed by the Chinese medicine high-dose group (hypoglycemic) and western medicine control group (U-mAlb lower) The content of GLUT1 was the highest in the serum of rats, and the content of GLUT1 was significantly decreased after treatment. The preventive effect of traditional Chinese medicine and the low dose of traditional Chinese medicine were the best, followed by the Western medicine control group. The pathological changes of renal tissue in each group were obviously lighter In the model group. 【Conclusion】 The effect of traditional Chinese medicine compound prescription for preventing and treating early diabetic nephropathy may be related to reducing the content of GLUT1 in serum.
