[目的]探讨大鼠急性颈脊髓损伤后黄芪多糖与CD44含量变化的关系。[方法]成年Wistar大鼠80只,体重240~250 g,雌雄不限。大鼠随机分为四组,分别为黄芪多糖组、生理盐水组、空白对照组、假手术组,每组20只。各组按取材时间不同又分为造模后24 h、3 d、1周、2周共4个时间点,每个时间点5只大鼠。采用颈7段脊髓改良Allen’s打击法制作大鼠脊髓损伤模型,打击力度为10×2.5g.cm;假手术组仅暴露颈7段脊髓。[结果]在不同时间点处死大鼠,测量各时间点每组大鼠血清和肺泡灌洗液中CD44含量。结果显示黄芪多糖对脊髓损伤大鼠CD44含量变化有影响。伤后3d黄芪多糖组血清及肺泡灌洗液CD44含量较空白对照组和生理盐水组有显著差异性(P<0.05)。[结论]黄芪多糖可以减轻大鼠急性颈脊髓损伤后血清CD44下降幅度并提高伤后3d肺泡灌洗液CD44含量。这一结果可能对治疗急性脊髓损伤后肺水肿有帮助。
[Objective] To investigate the relationship between the changes of astragalus polysaccharide and CD44 content after acute cervical spinal cord injury in rats. [Methods] 80 adult Wistar rats, weighing 240-250 g, were male and female. Rats were randomly divided into four groups, namely Astragalus polysaccharide group, saline group, blank control group and sham operation group, 20 rats in each group. Each group was divided into 4 h, 3 d, 1 wk, 2 wk, 4 wk, 5 rats at each time point. The model of spinal cord injury in rats was made by modified 7-segment spinal cord injury model of Allen’s. The striking force was 10 × 2.5g.cm. The sham-operated group only exposed the 7th spinal cord. [Result] The rats were sacrificed at different time points, and the contents of CD44 in serum and alveolar lavage fluid of each group were measured at each time point. The results showed that APS affected the content of CD44 in spinal cord injury rats. The contents of CD44 in serum and BALF of Astragalus polysaccharide group after 3d were significantly different from those in blank control group and saline group (P <0.05). [Conclusion] Astragalus polysaccharide can reduce the decrease of serum CD44 level and increase the content of CD44 in bronchoalveolar lavage liquid after acute cervical spinal cord injury in rats. This result may be helpful in the treatment of pulmonary edema after acute spinal cord injury.
