【目的】观察陈氏内异丸对子宫内膜异位症(endometriosis,EMs)大鼠异位内膜基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达的影响,探讨陈氏内异丸治疗EMs的作用机制。【方法】选用SD大鼠,随机分为假手术组,模型组,陈氏内异丸高、中、低剂量组(中药高、中、低剂量组,剂量分别为7.2、3.6、1.8 g.kg-1.d-1),丹那唑组(剂量为36 mg.kg-1.d-1)。复制EMs大鼠模型,造模成功后各组分别给予陈氏内异丸高、中、低剂量药液及丹那唑混悬液、蒸馏水等灌胃治疗4周。末次给药1周后剥取大鼠内异病灶组织检测MMP-2、MMP-9在异位内膜的表达。【结果】模型组大鼠MMP-2及MMP-9表达强度显著高于中药各剂量组和丹那唑组,差异均有统计学意义(P<0.01);中药高剂量组MMP-2及MMP-9表达强度显著低于中药中、低剂量组及丹那唑组,差异有统计学意义(P<0.05或P<0.01);中药中、低剂量组与丹那唑组比较差异均无统计学意义(P>0.05)。【结论】陈氏内异丸治疗EMs的作用与其能有效抑制大鼠异位内膜组织异常升高的MMP-2和MMP-9的表达有关。
【Objective】 To observe the effect of Chen’s Neiwan Pill on the expression of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in endometriosis (EMs) Affect the mechanism of Chen’s Neiwan Pill in the treatment of EMs. 【Methods】 SD rats were randomly divided into sham operation group, model group and Chen’s Nei Yi Wan high, medium and low dose groups (high, medium and low dose groups, the dosage was 7.2, 3.6, 1.8 g. kg-1.d-1), and the Danazol group (at a dose of 36 mg.kg-1.d-1). The model of EMs was duplicated. After the model was established successfully, the rats in each group were given high, medium and low dosage of Chen’s Nephritis pill, suspension of danazol, and distilled water for 4 weeks respectively. One week after the last administration, the endometriosis tissues were stripped to detect the expression of MMP-2 and MMP-9 in the ectopic endometrium. 【Results】 The expression of MMP-2 and MMP-9 in model group was significantly higher than that in traditional Chinese medicine and danazol groups (P <0.01). The MMP-2 and MMP- 9 expression was significantly lower than the traditional Chinese medicine, low-dose group and the Danazol group, the difference was statistically significant (P <0.05 or P <0.01); Chinese medicine, low-dose group and the Danazol group differences were not statistically significant P> 0.05). 【Conclusions】 The effect of Chen’s Neiwan Pill on EMs is related to its ability to inhibit the abnormally elevated expression of MMP-2 and MMP-9 in rat ectopic endometrium.
