目的原位检测肝细胞癌(HCC)中CD44v6mRNA及nm23H1mRNA的表达,了解CD44v6mRNA及nm23H1mRNA的表达与HCC侵袭转移的关系,了解HCC中CD44v6mRNA表达与nm23H1mRNA表达的相关性.方法采用PCR法合成CD44v6cDNA探针,体外转录法合成nm23H1cRNA探针,采用原位杂交法检测HCC33例CD44v6mRNA和nm23H1mRNA的表达.结果侵袭转移倾向高危组的10例HCC标本中,CD44v6mRNA和nm23H1mRNA的阳性率分别为800%(8/10)和400%(4/10);侵袭转移倾向低危组的23例HCC标本中,CD44v6mRNA和nm23H1mRNA的阳性率分别为217%(5/23)和913%(21/23).CD44v6mRNA的表达与HCC的侵袭转移倾向具有正相关性(P<001),nm23H1mRNA的表达与HCC的侵袭转移倾向具有负相关性(P<001),CD44v6mRNA及nm23H1mRNA的表达具有负相关性(P<001).结论检测CD44v6mRNA及nm23H1mRNA的表达可能成为HCC转移预后?
Objective To detect the expression of CD44v6 mRNA and nm23H1 mRNA in hepatocellular carcinoma (HCC) in situ, and to understand the relationship between the expression of CD44v6 mRNA and nm23H1 mRNA and the invasion and metastasis of HCC, and to understand the correlation between the expression of CD44v6 mRNA and the expression of nm23H1 mRNA in HCC. Methods The CD44v6 cDNA probe was synthesized by PCR and the nm23H1cRNA probe was synthesized by in vitro transcription method. The expression of CD44v6 mRNA and nm23H1 mRNA in 33 cases of HCC were detected by in situ hybridization. Results The positive rates of CD44v6 mRNA and nm23H1 mRNA in 10 HCC specimens with high risk of invasion and metastasis were 800% (8/10) and 400% (4/10), respectively; In 23 specimens of HCC, the positive rates of CD44v6 mRNA and nm23-H1 mRNA were 217% (5/23) and 913% (21/23), respectively. There was a positive correlation between the expression of CD44v6 mRNA and the invasion and metastasis tendency of HCC (P<001). The expression of nm23H1 mRNA was negatively correlated with the invasion and metastasis tendency of HCC (P<001), and the expression of CD44v6 mRNA and nm23H1 mRNA. The expression has a negative correlation (P<001). Conclusion The detection of CD44v6 mRNA and nm23-H1 mRNA may be the prognosis of HCC metastasis.
