目的:探讨吸烟与口腔黏膜上皮细胞NOD1信号通路关键分子表达水平的关系。方法:采用商业化可购的直接烟暴露装置,构建体外Leuk-1细胞吸烟模型,将其分为吸烟组(分别吸入香烟烟雾5、10、20、40、60 min)和对照组(吸入含5%CO2空气10 min),应用间接免疫荧光法检测体外吸烟对口腔黏膜上皮细胞NOD1通路内关键蛋白的表达。结果:随着体外吸烟时间的延长,NOD1蛋白表达水平逐渐下降,Caspase12和RIP2蛋白表达水平逐渐上升。NF-κB的蛋白表达水平逐渐上升之后明显下降。结论:吸烟可使口腔黏膜细胞NOD1信号通路相关蛋白表达发生改变,进而影响口腔黏膜固有免疫
Objective: To investigate the relationship between smoking and the expression of NOD1 signaling pathway key molecules in oral mucosa epithelial cells. Methods: Commercially available direct exposure smoke devices were used to establish Leuk-1 cell smoking model in vitro and divided into smoking groups (5, 10, 20, 40 and 60 min respectively) and control group 5% CO2 air for 10 min). The expression of key proteins in NOD1 pathway in oral mucosal epithelial cells was detected by indirect immunofluorescence. Results: NOD1 protein expression gradually decreased and the expression of Caspase12 and RIP2 protein gradually increased with the increase of smoking time in vitro. NF-κB protein expression levels gradually increased after the decline. Conclusion: Smoking can change the expression of NOD1 signal pathway related proteins in oral mucosal cells, and then affect the innate immunity of oral mucosa
