目的研究大鼠颈静脉回输自由基损伤红细胞所致的脑微血管功能障碍及硒的保护作用。方法采用Fenton羟自由基体系Haber-Weiss反应体外造成红细胞损伤 ,激光衍射法测定红细胞变形性表示其损伤程度。应用荧光标记和微循环数字图像处理技术分别检测对照组、自由基损伤组和硒保护组脑微血管管径、微血管自律运动、红细胞流速及微血管通透性的变化。结果自由基损伤导致红细胞变形性降低35 % ,硒保护组红细胞变形性仅降低15 %。脑微循环检测显示自由基损伤组微血管管径变小。微动脉管径变化较明显 ,5min达高峰 ,约30min恢复正常水平 ;微静脉管径变化较缓慢 ,10min达高峰 ,约60min恢复 ;微血管自律运动紊乱 ,微动脉较明显 ;红细胞流速明显减慢 ,5min时微动脉、微静脉内红细胞速度分别减慢31 %和35 % ;微血管通透性增加 ,5min时微动脉、微静脉通透性分别增加25 %和34 %。硒保护组上述指标均无显著性变化(P>0.05)。结论自由基损伤的红细胞可引起大鼠脑微血管功能障碍 ,微量元素硒有一定保护作用
Objective To investigate the protective effect of selenium on brain microvascular dysfunction induced by free radical-injured red blood cells in the jugular vein of rats. Methods Erythrocyte injury was induced by Fenton hydroxyl free radical system Haber-Weiss reaction in vitro. The degree of erythrocyte degeneration was measured by laser diffraction. Fluorescent labeling and microcirculation digital image processing techniques were used to detect the changes of cerebral microvessel diameter, autonomic activity of microvessels, erythrocyte flow rate and microvascular permeability in the control group, the free radical injury group and the selenium protection group respectively. Results Free radical damage resulted in a decrease of erythrocyte deformability of 35% and a decrease of 15% in selenoprotective erythrocyte deformability. Brain microcirculation tests showed that the diameter of microvessels in free radical injury group became smaller. The diameter of arterioles changed obviously, reached the peak at 5 min and returned to normal level at about 30 min. The diameter of venules changed slowly, reached the peak at 10 min and recovered at about 60 min. Microvascular autonomic dyskinesia and arterioles were more obvious. At 5 min, the arterioles and venules in the venules slowed by 31% and 35%, respectively. The microvascular permeability increased by 25% and 34% respectively at 5 minutes. Selenium protection group had no significant changes in the above indicators (P> 0.05). Conclusion Free radical injury of erythrocytes can cause cerebral microvascular dysfunction in rats and trace element selenium has some protective effect
