目的探讨吡哆醇(PN)对大鼠睾丸的体外毒性。方法采用在Wiliams方法的基础上改进的Sertoligerm细胞共培养系统,观察PN在不同剂量和接触时间对培养细胞的作用。结果脱落生精细胞数随PN浓度的增高和接触时间的延长而增加,并有明显的剂量—效应和时间—效应关系。同时,还观察到Sertoli细胞骨架出现松弛、回缩等效应。结论PN对大鼠生精细胞的体外效应反映了其对Sertoli细胞的损害。睾丸细胞共培养方法对探讨PN对大鼠睾丸的毒性作用具有实用价值。
Objective To investigate the in vitro toxicity of pyridoxine (PN) to testis in rats. Methods The improved Sertoli-germ co-culture system based on the Wiliams method was used to observe the effect of PN on cultured cells at different doses and contact time. Results The number of exfoliated spermatogenic cells increased with the increase of PN concentration and prolongation of exposure time with obvious dose-effect and time-effect relationship. At the same time, we also observed the relaxation, contraction and other effects of Sertoli cytoskeleton. Conclusions The in vitro effect of PN on rat spermatogenic cells reflects its damage to Sertoli cells. Testicular cell co-culture method to explore the toxic effects of PN on rat testes has practical value.
