试验采用正交试验设计优化猪肝中蛋白磷酸酶2A(Protein phosphatase 2A,PP2A)的提取工艺。以PP2A的活力为考察指标,在优化提取剂类型、pH、料液比和Triton-X100体积浓度四个单因素的基础上,再进行三因素三水平的正交试验。结果表明,最佳工艺条件:提取剂为含蔗糖的Tris缓冲液、pH为8.0、料液比为1∶4 g/mL、Triton-X100体积浓度为0.5%。在此条件下,总酶活为10.89 U。试验的开展将为PP2A的制备提供了一条新的途径。
Orthogonal design was used to optimize the extraction process of protein phosphatase 2A (PP2A) in porcine liver. The activity of PP2A was taken as the index, and three factors and three levels of orthogonal experiment were carried out on the basis of four single factor factors: extractant type, pH, solid-liquid ratio and volume concentration of Triton-X100. The results showed that the optimum conditions were as follows: the extractant was sucrose-containing Tris buffer, pH 8.0, the ratio of solid to liquid was 1: 4 g / mL and the volume concentration of Triton-X100 was 0.5%. Under this condition, the total enzyme activity was 10.89 U. The test will provide a new way for the preparation of PP2A.
