目的:探讨护肝片对睡眠剥夺大鼠肝损伤的保护作用。方法:取SD大鼠72只,随机分成6组,每组12只,分别为:正常对照组、模型组、一贯煎阳性对照组及护肝片低、中、高剂量组。灌胃给药,1次/d,连续16 d,于给药第17天采用改良多平台睡眠剥夺法造模,连续造模9 d,每日剥夺20 h。于造模第9天给药30 min后,采血分离血清检测IL-6、IL-12、AST、ALT水平,称量肝脏湿质量,计算肝脏系数,取肝组织匀浆检测MDA、SOD、GSH水平,取肝切片,HE染色,光镜观察。结果:与模型组比较,护肝片可显著降低睡眠剥夺模型大鼠血清IL-6、IL-12、ALT及肝组织MDA水平,提高肝组织GSH、SOD水平(P<0.05);肝组织病理学检查结果显示护肝片对睡眠剥夺大鼠的肝损伤有一定程度的保护作用。结论:护肝片具有显著降低肝损伤的作用,其作用机理可能与提高机体免疫功能和抗氧化能力有关。
Objective: To explore the protective effect of Hugan Tablets on liver injury in rats after sleep deprivation. Methods: Seventy-two SD rats were randomly divided into 6 groups (12 rats in each group), which were normal control group, model group, usual Jiangyang control group and Hugan tablet low, medium and high dose groups respectively. Gavage once a day for 16 days. On the 17th day after administration, a modified multi-platform sleep deprivation method was used to establish the model. The model was continuous for 9 days and deprived for 20 hours daily. Blood samples were collected for determination of IL-6, IL-12, AST and ALT after 30 min of administration on the 9th day of modeling. The liver wet weight was measured and the liver coefficient was calculated. The contents of MDA, SOD and GSH Levels, liver slices, HE staining, light microscopy. Results: Compared with the model group, Hugan tablet can significantly reduce the level of serum IL-6, IL-12, ALT and MDA in liver tissue and increase the levels of GSH and SOD in liver tissue (P <0.05) Neutropenia test showed that Hugan tablet can protect the liver from sleep deprived rats to a certain extent. Conclusion: Hugan Tablets can significantly reduce hepatic injury and its mechanism may be related to the improvement of immune function and antioxidant capacity.
