目的:通过实验观察龟甲胶、鹿角胶含药血清对豚鼠膝骨关节炎软骨细胞促增殖作用以及两种含药血清对JNK及p38 MAPK基因表达的影响,揭示龟甲胶及鹿角胶治疗膝骨关节炎的作用机制。方法:分别用龟甲胶、鹿角胶、盐酸氨基葡萄糖(GHC)含药血清及生理盐水对3月龄豚鼠骨关节炎软骨细胞进行干预,MTT法比较含药血清对软骨细胞的促增殖情况,QPCR检测含药血清干预后软骨细胞JNK及p38 MAPK基因的表达情况。结果:用含药血清浓度为5%的各组培养液干预后(时间72h),龟甲胶组、鹿角胶组、氨基葡萄糖组和空白对照组MTT法检测OD值分别为0.468±0.017,0.425±0.010,0.337±0.017,0.276±0.027,结果显示差异有统计学意义(P<0.05)。QPCR检测龟甲胶组、鹿角胶组、盐酸氨基葡萄糖组和空白对照组JNK mRNA相对表达量分别为0.162±0.096,0.333±0.165,0.653±0.141,1.000±0.000,差异有统计学意义(P<0.05)。QPCR检测龟甲胶组、鹿角胶组、盐酸氨基葡萄糖组和空白对照组p38MAPK mRNA相对表达量分别为0.058±0.025,0.229±0.057,0.441±0.090,1.000±0.000,结果显示差异有统计学意义(P<0.05)。结论:龟甲胶和鹿角胶都能有效减少豚鼠骨关节炎软骨细胞JNK及p38 MAPK基因的表达,对软骨细胞作用为促增殖,对其凋亡产生抑制作用,从而一定程度上减缓骨关节炎的病损进展。
OBJECTIVE: To observe the effects of TCDA and antler on the proliferation of chondrocytes of knee osteoarthritis in guinea pigs and the effects of two serums on the expression of JNK and p38 MAPK, and to reveal the effects of Tortala and antler on knee joint The mechanism of inflammation. Methods: Osteoarthritis chondrocytes of 3-month guinea pigs were intervened by using Tortala, guar gum, glucosamine hydrochloride (GHC) -containing serum and normal saline respectively. The proliferation of chondrocytes was assayed by MTT assay. Detect the expression of JNK and p38 MAPK gene in chondrocytes after intervention with drug serum. Results: The MTT assay showed that the OD value of each group was 0.468 ± 0.017 and 0.425 ± (P <0.05) after intervention with 5% serum concentration for 72 h, TQA group, antler gum group, glucosamine group and blank control group 0.010,0.337 ± 0.017,0.276 ± 0.027, the results showed that the difference was statistically significant (P <0.05). The relative expression levels of JNK mRNA in QCM test group were 0.162 ± 0.096, 0.333 ± 0.165, 0.653 ± 0.141 and 1.000 ± 0.000, respectively, with significant difference (P <0.05) ). The relative expression levels of p38MAPK mRNA in QCM test group, TGP group, glucosamine hydrochloride group and blank control group were 0.058 ± 0.025,0.229 ± 0.057,0.441 ± 0.090,1.000 ± 0.000, respectively. The results showed that the difference was statistically significant (P <0.05). CONCLUSION: Tortala and antler can effectively reduce the expression of JNK and p38 MAPK in chondrocytes of osteoarthritis of guinea pigs, and promote the proliferation of chondrocytes, which can inhibit the apoptosis of osteochondritis cells and thus reduce the degree of osteoarthritis Lesion progression.
