异硫氰酸-2,6-二甲基苯酯(1)是合成镇痛药赛拉嗦(Xylazine)的主要中间体。[Brit 1001091;(Ger1173475;CA 1964,61:13323c)]。工业生产用2,6-二甲基苯胺和硫氰酸铵,在氯化氢存在下,于氯苯中回流反应制得,[Brit 1099768;(CA 1968,69:18823e)]。该方法原料易得,具有一定的实用价值。但其缺点是HCl严重腐蚀设备,并有劳动保护问题。作者经实验,以浓硫酸代替HCl,后处理并以石油醚代替正已烷得到满意结果,收率62%。改进后操作简便,成本节省,具有实用价值。产品经鉴定与文献报道一致。
2,6-Dimethylphenyl isothiocyanate (1) is the main intermediate for the synthetic analgesic Xylazine. [Brit 1001091; (Ger 1173475; CA 1964, 61: 13323c)]. Commercial production of 2,6-dimethylaniline and ammonium thiocyanate in the presence of hydrogen chloride, refluxing in chlorobenzene prepared [Brit 1099768; (CA 1968,69: 18823e)]. The method is easy to get raw materials, has a certain practical value. However, the disadvantage is that HCl severely corrodes the equipment and has labor protection problems. The experiment by concentrated sulfuric acid instead of HCl, post-processing and replacement of n-hexane with petroleum ether to obtain satisfactory results, yield 62%. Improved after the operation is simple, cost savings, with practical value. Products identified and reported in the literature.
