目的探讨鸟苷酸交换因子C3G(Crk SH3-domain-binding guanine nucleotide-releasing factor)在卵巢癌组织中的表达及在卵巢癌SKOV3细胞增殖中的作用。方法采用Western blot检测比较40例卵巢癌、8例卵巢良性肿瘤及4例正常卵巢组织中C3G的表达水平。应用特异性siRNA下调SKOV3细胞中C3G的表达,通过MTT法和平板克隆实验检测细胞增殖,建立裸鼠皮下种植瘤模型观察移植瘤生长情况。结果卵巢癌组织中C3G的表达显著高于卵巢良性肿瘤及正常卵巢组织,其在卵巢癌、卵巢良性肿瘤分别是正常卵巢组织中的(1.832±0.200)(P<0.05)、(0.893±0.147)倍。siRNA干扰可使SKOV3细胞中内源性C3G的表达下调79.2%;与野生型细胞组、阴性对照组及空载体对照组相比,C3G干扰组细胞增殖变慢(P<0.05),克隆形成率下降(P<0.05),移植瘤的平均体积和质量显著下降(P<0.05)。结论鸟苷酸交换因子C3G在大多数上皮性卵巢癌中过表达,并促进卵巢癌细胞增殖。
Objective To investigate the expression of Crk SH3-domain-binding guanine nucleotide-releasing factor (OVX) in ovarian cancer and its role in the proliferation of ovarian cancer SKOV3 cells. Methods Western blot was used to detect the expression of C3G in 40 cases of ovarian cancer, 8 cases of benign ovarian tumor and 4 cases of normal ovarian tissue. The specific siRNA was used to down-regulate the expression of C3G in SKOV3 cells. The proliferation of SKOV3 cells was detected by MTT assay and cell cloning assay. The growth of xenografts in nude mice was observed. Results The expression of C3G in ovarian cancer tissues was significantly higher than that in benign ovarian tumors and normal ovarian tissues (1.832 ± 0.200), (0.893 ± 0.147) in ovarian cancer and benign ovarian tumor, respectively Times siRNA interference could down-regulate the expression of endogenous C3G in SKOV3 cells by 79.2%. Compared with the wild-type cell group, negative control group and empty vector control group, the cell proliferation of C3G interference group was slower (P <0.05) (P <0.05), and the average volume and quality of xenografts decreased significantly (P <0.05). Conclusion The guanylate exchange factor C3G is overexpressed in most epithelial ovarian carcinomas and promotes the proliferation of ovarian cancer cells.
