目的探讨2,4-D丁酯(butyl 2,4-dichlorophenoxyacetate,2,4-D b.e.)亚慢性染毒对雄性大鼠脑组织中氨基酸类神经递质谷氨酸(Glu)和γ-氨基丁酸(GABA)含量的影响。方法将40只健康30 d龄SPF级雄性Wistar大鼠随机分为4组,分别为对照(玉米油:蒸馏水体积比为1:1)组和低(1/100 LD_(50),6 mg/kg)、中(3/100 LD_(50),18 mg/kg)和高剂量(6/100 LD_(50),36 mg/kg)2,4-D丁酯染毒组,每组10只。经口灌胃染毒,每周连续染毒5 d,实验周期为90 d。染毒结束后,高效液相色谱法检测大鼠大脑皮层、海马、小脑中GABA、Glu的含量。结果随着2,4-D丁酯染毒剂量的升高,大鼠大脑皮层、海马、小脑中的Glu含量和Glu/GABA值均呈升高趋势;大脑皮层和海马中的GABA含量呈下降趋势,小脑中GABA含量呈先上升后下降的趋势。结论 2,4-D丁酯亚慢性染毒可破坏小鼠脑组织中Glu和GABA水平之间的动态平衡。
Objective To investigate the effects of 2,4-D bezoar (2,4-D be) on the expression of glutamate (Glu) and γ-amino Butyric acid (GABA) content. Methods Forty healthy male Sprague-Dawley (Wistar) rats, aged 30 days, were randomly divided into four groups: control (corn oil: distilled water volume ratio of 1: 1) group and low (1/100 LD 50, 6 mg / kg), medium (3/100 LD_ (50), 18 mg / kg) and high dose (6/100 LD_ (50), 36 mg / kg) . Oral administration of oral administration, weekly continuous exposure to 5 d, the experimental period of 90 d. At the end of exposure, the contents of GABA and Glu in the cerebral cortex, hippocampus and cerebellum of rats were determined by high performance liquid chromatography. Results Glu levels and Glu / GABA values in cerebral cortex, hippocampus and cerebellum increased with the increasing doses of 2,4-D butyl ester in rats. The levels of GABA in cerebral cortex and hippocampus decreased Trend, the content of GABA in cerebellum first increased and then decreased. Conclusion Subchronic exposure to 2,4-D butyl can destroy the dynamic balance between Glu and GABA levels in mouse brain.
