目的研究大黄酸对镉致雄性大鼠生殖毒性的干预作用。方法雄性SD大鼠32只,随机分为四组,分别为空白对照组、染镉组、大黄酸低剂量组和大黄酸高剂量组。除空白组外其余各组大鼠腹腔注射氯化镉染毒,每2d 1次,染毒10次,染毒的同时灌胃给药,1次/d,连续给药6周。给药结束后,测定大鼠附睾精子数,测定睾丸组织中超氧化物歧化酶(SOD)和半胱氨酸天冬氨酸蛋白酶(caspase-3)的活力,测定丙二醛(MDA)的水平,HE染色法观察睾丸组织病理变化。结果与空白对照组比,染镉组大鼠精子数量和精子活动率下降;SOD活力明显降低;精子畸形率、MDA和caspase-3水平升高(P<0.05)。大黄酸给药组精子数量、活动率和SOD活力升高,精子畸形率MDA和caspase-3水平降低;病理切片显示:染镉组睾丸生精小管被破坏,生精细胞数目减少,大黄酸高剂量组大鼠生精小管基本完整。结论大黄酸对镉造成的雄性大鼠生殖毒性有明显的干预作用。
Objective To study the effect of rhein on the reproductive toxicity of male rats induced by cadmium. Methods Thirty-two male SD rats were randomly divided into four groups: blank control group, cadmium-exposed group, low-dose rhein group and high-dose rhein group. Except the blank group, the other rats in each group were intraperitoneally injected with cadmium chloride, once every 2 days for 10 times. The rats were orally administered once a day for 6 weeks. After administration, the number of epididymal sperm in rats was determined, the activities of superoxide dismutase (SOD) and caspase-3 in testis tissue were determined, and the level of malondialdehyde (MDA) The pathological changes of testis were observed by HE staining. Results Compared with the blank control group, the sperm count and sperm motility of cadmium-exposed rats decreased significantly; SOD activity decreased significantly; sperm deformity rate, MDA and caspase-3 levels increased (P <0.05). Rhein administration group sperm quantity, activity rate and SOD activity increased, sperm deformity rate MDA and caspase-3 levels decreased; pathological sections showed: cadmium testicular seminiferous tubules were destroyed, the number of spermatogenic cells decreased, rhein high The spermatogonium in the dose group was basically intact. Conclusion Rhein has significant effect on the reproductive toxicity of male rats caused by cadmium.
