目的构建豚鼠颈内静脉置管模型,解决豚鼠静脉多次、大剂量给药问题。方法 10只豚鼠麻醉后均在显微镜下切开颈内静脉,置入医用PE导管,术后7d每日换药并向导管内注射1mL肝素化的生理盐水(50U/mL),观察有无堵管、脱落现象以及伤口有无红肿、渗液,术后第7d抽取静脉血行血常规及“C”反应蛋白检查。结果①术后观察7d,10只豚鼠存活8只,2只豚鼠于第4d死亡,每日观察无脱管及堵管现象。②术后第1d2只豚鼠伤口有明显渗出,略肿胀,第2d渗出减轻,无肿胀,此后至第7d伤口无渗出及肿胀出现;③术前与术后7d血常规主要指标与“C”反应蛋白均无明显差异。结论成功构建豚鼠颈内静脉置管模型,可满足静脉多次、大剂量给药的需要。
Objective To construct a model of cannulation of guinea pig internal jugular vein to solve the problem of multiple and high doses of intravenous injection in guinea pigs. Methods Ten guinea pigs were anesthetized and their internal jugular vein was dissected under the microscope. The medical PE catheter was placed on the catheter. After 7 days of operation, the drug was changed daily and 1 mL of heparinized normal saline (50 U / mL) , The phenomenon of shedding and whether the wounds were swollen and exudate, blood samples were taken from the 7th day after operation to examine the blood and blood samples for “C” reaction. Results ① After operation for 7 days, only 10 guinea pigs survived, and 2 guinea pigs died on the 4th day. No detachment and occlusion were observed daily. ② The first day after operation, the guinea pigs showed obvious exudation and slightly swollen on the 1st day after operation. The exudation on the 2nd day was relieved and no swelling was found. No exudation and swelling occurred on the 7th day after the operation. The main indexes of blood routine at 7 days before and after surgery were “C ” reactive protein were no significant differences. Conclusion The successful establishment of guinea pig jugular vein catheterization model can meet the needs of intravenous multiple, high-dose administration.
