目的探讨骨形态发生蛋白-7(bone morphogenetic protein-7,BMP-7)对马兜铃酸(Aristolchic acid,AA)诱导的肾小管上皮细胞转分化的拮抗作用,以期为马兜铃酸肾病(Aristolchic acid nephropathy,AAN)寻找治疗新措施。方法应用不同浓度BMP-7(75ng/mL、150ng/mL、300ng/mL)处理经AA(10μg/mL)诱导转分化的体外培养人近端肾小管上皮细胞株(HK-2细胞),然后应用逆转录-聚合酶链反应(RT-PCR)及Western blotting技术分别检测α-SMA mRNA及蛋白的表达水平。结果BMP-7呈剂量依赖方式逆转AA诱导的HK-2细胞α-SMA基因及蛋白的表达。结论BMP-7可以通过抑制马兜铃酸诱导的人肾小管上皮细胞转分化从而可能成为AAN的防治潜在新药。
Objective To investigate the antagonistic effect of bone morphogenetic protein-7 (BMP-7) on the transdifferentiation of renal tubular epithelial cells induced by aristolochic acid (AA) in order to provide a basis for aristolochic acid nephropathy Aristolchic acid nephropathy, AAN) looking for new treatments. Methods Human proximal tubular epithelial cell lines (HK-2 cells) were induced by AA (10μg / mL) with different concentrations of BMP-7 (75ng / mL, 150ng / mL and 300ng / mL) The expression of α-SMA mRNA and protein were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting respectively. Results BMP-7 reversed the AA-induced α-SMA gene and protein expression in HK-2 cells in a dose-dependent manner. Conclusion BMP-7 may be a potential new drug for AAN by inhibiting aristolochic acid-induced human renal tubular epithelial cell transdifferentiation.
