目的 探讨环孢素A(CsA)慢性肾毒性的发生机理。方法 给予进正常饮食或低盐饮食的大鼠灌服CsA 3 0mg·kg-1·d-1,采用逆转录聚合酶链反应 (RT PCR)研究CsA对大鼠肾内转化生长因子 β1(TGF β1)及肾素mRNA表达的影响。 结果 给予CsA的大鼠均有肾内TGF β1mRNA、肾素mRNA表达的增加 ;给予低盐饮食的大鼠这些改变更为明显。结论 CsA的慢性肾毒性与肾内TGF β1mRNA、肾素mRNA表达的增加有关。
Objective To investigate the mechanism of chronic nephrotoxicity induced by cyclosporine A (CsA). Methods CsA 3 0 mg · kg-1 · d-1 was administrated into normal diet or low-salt diet, and the effect of CsA on the expression of transforming growth factor-β1 (TGFβ1) in rat kidney was analyzed by reverse transcriptase-polymerase chain reaction β1) and renin mRNA expression. Results All CsA-treated rats had increased TGFβ1 mRNA and renin mRNA expression in the kidney. These changes were more pronounced in rats given low-salt diet. Conclusion The chronic nephrotoxicity of CsA is related to the increase of renal TGFβ1 mRNA and renin mRNA expression.
