目的:探讨二烯丙基二硫(DADS)对白血病K562细胞增殖的影响,以及Bcl-2表达的调节作用。方法:用DADS预处理白血病K562细胞构建细胞模型,MTT法分别检测不同DADS浓度(5gmL-1、10gmL-1、20gmL-1、40gmL-1)和不同处理时间(6h、12h、24h、48h)细胞增殖情况,IC50浓度处理白血病K562细胞之后,Westernblot和RT-PCR检测Bcl-2蛋白和mRNA表达水平。结果:MTT结果显示DADS能够抑制白血病K562细胞的增殖,且呈剂量和时间依赖性;IC50浓度的DADS处理K562细胞24小时后,Bcl-2蛋白和mRNA的表达量显著减少。结论:DADS可显著抑制K562细胞增殖,而这一作用可能与Bcl-2表达下调有关。
Objective: To investigate the effects of diallyl disulfide (DADS) on the proliferation of leukemia K562 cells and the regulation of Bcl-2 expression. Methods: The cell model of leukemia K562 cells was pretreated with DADS. MTT assay was used to detect the expression of DADS in different concentrations of DADS (5g mL-1,10g mL-1,20g mL-1,40g mL-1) and different treatment time (6h, 12h, 24h, 48h) Cell Proliferation, IC50 Concentration After leukemia K562 cells were treated, the expression of Bcl-2 protein and mRNA were detected by Western blot and RT-PCR. Results: The results of MTT showed that DADS could inhibit the proliferation of K562 cells in a dose-and time-dependent manner. The expression of Bcl-2 protein and mRNA in K562 cells treated with IC50 for 24 hours was significantly decreased. Conclusion: DADS can significantly inhibit the proliferation of K562 cells, which may be related to the down-regulation of Bcl-2 expression.
