1971年我们制成右旋糖酐-泛影葡胺液分离血中淋巴细胞取得满意效果。随后用Ficcoll-Hypaque和聚甘露醇-泛影葡胺也取得同样效果。脱纤维使血的粒细胞显著降低,本文报告无空调试验室在深秋低温10~15℃和盛夏高温30~35℃时分离淋巴细胞的技术。 我们设计了控制离心筒内微环境的温度,深秋时将离心筒预热至25℃,盛夏时10℃冰箱预冷2小时,再加脱纤维血于分离液面,离心后可得满意的分离效果。
In 1971 we made dextran - meglumine diacetate blood lymphocytes obtained satisfactory results. The same effect was then achieved with Ficcoll-Hypaque and polymannitol-diatrizoate. The defibrillating results in a significant reduction of blood granulocytes. In this paper, we report the technique of separating lymphocytes from air-conditioned laboratories at late autumn at 10-15 ℃ and midsummer at high temperatures at 30-35 ℃. We designed to control the microenvironment of the centrifuge tube temperature, the late autumn when the centrifuge tube preheated to 25 ℃, 10 ℃ in the early summer when the refrigerator precooling, coupled with defibrillation in the separation of blood, centrifuged to obtain a satisfactory separation effect.
