目的研究白藜芦醇对异氟醚吸入诱发老年小鼠认知功能障碍及海马组织炎症反应的影响。方法 48只15个月龄雄性C57BL/6小鼠随机均分为四组:A和B组连续7d腹腔注射生理盐水后分别吸入氧气和1.5%异氟醚2h;C和D组连续7d腹腔注射白藜芦醇100mg/kg后吸入氧气和1.5%异氟醚2h。气体吸入后24h行旷场实验和条件恐惧性实验;6h和48h取海马组织,采用ELISA或Western blot法检测IL-1β、TNF-α、NF-κB抑制蛋白α(IκBα)和Nod样受体家族包含pyrin结构域蛋白(NLRP3)水平。结果 B组环境诱发僵直时间百分比低于A、D组(P<0.05)。气体吸入后6h和48h,B组海马IL-1β、TNF-α和NLRP3水平高于A、D组(P<0.05),IκBα水平低于A、D组(P<0.05)。结论白藜芦醇可改善异氟醚吸入诱发的小鼠认知功能障碍,可能与其减轻海马组织炎症反应有关。
Objective To investigate the effects of resveratrol on cognitive dysfunction and hippocampal inflammatory response induced by isoflurane inhalation in aged mice. Methods Forty-eight male C57BL / 6 mice, 15 months old, were randomly divided into four groups: group A and group B were treated with intraperitoneal injection of normal saline for 7 days and oxygenated with 1.5% isoflurane for 2 hours respectively; group C and D were injected intraperitoneally Resveratrol 100 mg / kg inhaled oxygen and 1.5% isoflurane 2h. Open field test and conditional fear test were performed 24h after gas inhalation. Hippocampus were harvested at 6h and 48h. The expressions of IL-1β, TNF-α, NF-κB inhibitor IκBα and Nod-like receptor Family contains pyrin domain protein (NLRP3) levels. Results The percentage of environment-induced stiffness in group B was lower than that in group A and D (P <0.05). The levels of IL-1β, TNF-α and NLRP3 in hippocampus of group B were higher than those in groups A and D at 6h and 48h after inhalation of gas (P <0.05). The levels of IκBα in group B were lower than those in groups A and D (P <0.05). Conclusion Resveratrol can improve cognitive impairment in mice induced by inhalation of isoflurane, which may be related to the reduction of inflammation in hippocampus.
