目的探讨新疆柯尔克孜族人群人类血小板抗原HPA1-6,15(Gov)系统多态性,分析HPA基因在柯尔克孜族人群中的遗传多态性。方法采用聚合酶链-序列特异性引物(PCR-SSP)法105例无血缘关系的新疆柯尔克孜族血样进行HPA基因分型。结果新疆柯尔克孜族HPA-1a、2a、3a、4a、5a、6a、15a基因频率分别是0.900 0、0.923 80、.576 2、1、0.952 4、10、.566 7;HPA-1b、2b、3b、4b、5b、6b、15b基因频率分别是0.100 00、.076 2、0.423 8、0、0.047 6、00、.433 3,新疆柯尔克孜族HPA基因频率与新疆汉族相比,HPA-1a、1b、2a、2b、15a、15b基因频率差异具有统计学意义(P<0.05)。与新疆维吾尔族相比,HPA-15a、15b基因频率差异具有统计学意义(P<0.05)。结论新疆柯尔克孜族人群中HPA-1、2、3、5、15系统均具有多态性,其中HPA-3,15系统具有高度多态性,这使免疫暴露的机会增加。
Objective To investigate the polymorphism of human platelet antigen HPA1-6,15 (Gov) in Kirgiz population in Xinjiang and to analyze the genetic polymorphism of HPA gene in Kirgiz population. Methods Polymerase chain-specific primers (PCR-SSP) were used to detect HPA genotypes in 105 blood samples of unrelated Xinjiang Kirgiz. Results The gene frequencies of HPA-1a, 2a, 3a, 4a, 5a, 6a and 15a in the Kirgiz population in Xinjiang were 0.900 0,0.923 80, .576 2,1,0.952 4,10 and .566 7, respectively. HPA-1b, 3b, 4b, 5b, 6b, 15b gene frequencies were 0.100 00, .076 2,0.423 8,0,0.047 6,00, .433 3, Xinjiang Kirgiz family HPA gene frequency compared with Han Chinese, HPA-1a, 1b, 2a, 2b, 15a, 15b were significantly different (P <0.05). Compared with Xinjiang Uygur, the differences of HPA-15a and 15b gene frequencies were statistically significant (P <0.05). Conclusion The HPA-1, 2, 3, 5 and 15 systems in Xinjiang Kirgiz population are all polymorphic. Among them, the HPA-3 and 15 systems are highly polymorphic, which increases the chance of immune exposure.
