采用液体反相悬浮法制取壳聚糖微球,在微球表面偶联纳豆激酶的亲和配体—大豆蛋白,作为纳豆溶栓酶的亲和吸附介质。结果表明:壳聚糖微球本身对纳豆激酶没有特异吸附,偶联大豆蛋白的壳聚糖微球对发酵粗酶液中的纳豆激酶的吸附性能符合Langmuir方程,吸附平衡时间约为60 min,最大吸附量为3 926.56 U·g-1。纳豆激酶发酵液的上样量与柱体积比为1∶9时,纳豆激酶的收率可以达到52.3%,纯化倍数可以达到18.1倍。
Chitosan microspheres were prepared by liquid reversed-phase suspension method, and the affinity ligand-nattokinase of nattokinase was coupled on the surface of microspheres to act as the affinity adsorbent for natto-thrombolytic enzyme. The results showed that the chitosan microspheres did not specifically adsorb natto kinase. The adsorption properties of the conjugate soy protein chitosan microspheres to the nattokinase in the crude fermentation broth were in accordance with the Langmuir equation. The adsorption equilibrium time was about 60 min, the maximum adsorption capacity is 3 926.56 U · g-1. Nattokinase fermentation broth volume and column volume ratio of 1: 9, natto kinase yield can reach 52.3%, purification fold can reach 18.1 times.
