姜黄素是一种新的渔用饲料添加剂,但目前尚无测定鱼饲料中姜黄素类化合物添加量检测方法。本研究建立了鱼饲料中双去甲氧基姜黄素、去甲氧基姜黄素和姜黄素同时测定的超高效液相色谱紫外法(UPLC-TUV)。方法采用乙醇作第一提取剂,乙腈为第二提取剂,基质标准曲线定量。以乙腈-0.1%乙酸水溶液(45∶55,V/V)为流动相,C_(18)柱(2.1 mm×100 mm,1.7μm)为色谱柱,430 nm波长下进行检测。鱼饲料中双去甲氧基姜黄素、去甲氧基姜黄素和姜黄素最低检测限为10μg/kg,定量限为25μg/kg。在空白鱼饲料中添加双去甲氧基姜黄素、去甲氧基姜黄素和姜黄素的水平在0.10~2.0 mg/kg时,平均回收率为87.68%~102.06%,相对标准偏差为1.06%~8.01%。本方法为检测鱼饲料中双去甲氧基姜黄素、去甲氧基姜黄素和姜黄素的添加量提供了一种技术手段。
Curcumin is a new fishery feed additive, but there is no method to detect the amount of curcumin in fish feed. In this study, UPLC-TUV was established for the simultaneous determination of didemethoxycurcumin, demethoxycurcumin and curcumin in fish feed. Methods Ethanol was used as the first extractant, acetonitrile as the second extractant, and the matrix standard curve was quantified. The mobile phase was acetonitrile-0.1% aqueous acetic acid (45:55, V / V) and the column was C 18 column (2.1 mm × 100 mm, 1.7 μm). The lowest limit of detection of didemethoxy curcumin, demethoxycurcumin and curcumin in fish feed was 10 μg / kg and the limit of quantification was 25 μg / kg. The average recoveries of double demethoxycurcumin, demethoxycurcumin and curcumin at the blank feed of 0.10 to 2.0 mg / kg were 87.68% -102.06% with relative standard deviations of 1.06% ~ 8.01%. The method provides a technical measure for detecting the amount of didemethoxy curcumin, demethoxy curcumin and curcumin in the fish feed.
