目的探讨抑制肽酰基精氨酸脱亚氨酶4(PADI4)对卵巢癌细胞系OVCAR3的影响。方法 PADI4-si RNA组和阴性对照组si RNA分别转染卵巢癌细胞OVCAR3,采用实时定量PCR及Western Blotting检测PADI4、p53和p21的表达,AnnexinⅤ-PE/7-AAD双染和transw ell小室法分别检测细胞凋亡、侵袭和迁移的情况。结果与阴性对照组相比,PADI4-si RNA组的OVCAR3细胞中PADI4表达降低,且其凋亡细胞比例明显增加(P<0.05),细胞侵袭能力明显减弱(P<0.01),但细胞趋化能力没有明显变化。PADI4-si RNA组p53表达较阴性对照组降低,但p21表达并没有降低。结论沉默PADI4影响卵巢癌细胞OVCAR3凋亡和侵袭特性,提示PADI4参与调控卵巢癌细胞恶性生物学行为。
Objective To investigate the effect of inhibition of PADI4 on ovarian cancer cell line OVCAR3. Methods PADI4-si RNA and negative control siRNA were transfected into ovarian cancer cells OVCAR3 respectively. The expression of PADI4, p53 and p21 were detected by real-time quantitative PCR and Western Blotting. The AnnexinⅤ-PE / 7-AAD double staining and transw ell cell method The apoptosis, invasion and migration were detected respectively. Results Compared with the negative control group, the expression of PADI4 in OVCAR3 cells was decreased in PADI4-si RNA group and the percentage of apoptotic cells was significantly increased (P <0.05), while the invasion ability was significantly decreased (P <0.01) No significant change in ability. PADI4-si RNA group p53 expression decreased compared with the negative control group, but p21 expression did not decrease. Conclusion Silencing PADI4 can affect the apoptosis and invasion of ovarian cancer cells OVCAR3, suggesting that PADI4 is involved in the regulation of malignant biological behavior of ovarian cancer cells.
