目的研究人肺癌相关基因LSCC3表达与肺癌分期,病理类型,预后之间的关系。方法提取50对肺癌组织和癌旁正常肺组织标本的总核糖核酸(RNA),以人类小核糖体RNA(18srRNA)为内参照行半定量RNA反转录聚合酶链式反应(RTPCR),检测LSCC3在肺癌组织和癌旁正常肺组织中的差异表达,并结合50例患者的临床资料,分析该基因的表达程度与肺癌病理类型、pTNM分期、无瘤生存时间,生存率之间的关系。结果LSCC3基因在肺癌组织中的表达低于癌旁正常肺组织(P<0.01);该基因的表达程度与肺癌患者的性别(P>0.05)、病理类型(P>0.05)、肿瘤分化程度(P>0.05)、病理分期(P>0.05)之间无明显相关;肺癌组织中LSCC3基因高表达患者的无瘤生存时间和生存率高于LSCC3基因低表达患者。结论LSCC3可认为是一个肺癌相关基因,并且可能是一个抑癌基因;LSCC3基因高表达患者预后较低表达者明显改善。
Objective To study the relationship between the expression of lung cancer related gene (LSCC3) and the staging, pathological type and prognosis of lung cancer. Methods Total RNA was extracted from 50 lung cancer tissues and adjacent normal lung tissue samples. Semi-quantitative reverse transcriptase-polymerase chain reaction (RTPCR) was performed using small human ribosomal RNA (18srRNA) LSCC3 was differentially expressed in lung cancer tissues and adjacent normal lung tissues. The clinical data of 50 patients were analyzed to analyze the relationship between the expression of this gene and the pathological type, pTNM stage, tumor-free survival time and survival rate. Results The expression of LSCC3 in lung cancer tissues was lower than that in adjacent normal lung tissues (P <0.01). The expression level of LSCC3 gene in lung cancer patients was significantly higher than that in lung cancer patients (P> 0.05), pathological type (P> 0.05) (P> 0.05). There was no significant correlation between the pathological stage (P> 0.05) and the tumor-free survival time and survival rate of LSCC3 gene in patients with lung cancer. Conclusion LSCC3 is considered as a lung cancer-related gene and may be a tumor suppressor gene. LSCC3 gene expression in patients with low expression of the prognosis was significantly improved.
