目的观察生长抑素对人胰腺癌细胞株HS76 6T的生长抑制作用并探讨其作用机制。方法细胞培养 ,分别加入不同浓度的生长抑素 ,应用四甲基偶氮唑盐法观察细胞增殖程度 ;放射免疫法测定细胞内cAMP含量 ;Fura 2 AM测定细胞内Ca2 +浓度。结果不同浓度的生长抑素 (10 - 6~10 - 1 2 M)均能有效地抑制HS76 6T的生长 ,能抑制HS76 6T细胞内cAMP生成和Ca2 +水平 ,分别在 5min时和 3min时作用最大 ,cAMP生成量和Ca2 +水平与生长抑素浓度呈负相关。它能使垂体腺苷酸环化酶激活多肽 (PACAP)诱导的细胞内cAMP生成量和Ca2 +减少。结论生长抑素能抑制人胰腺癌HS76 6T细胞的生长 ,可明显抑制PACAP诱导的细胞的生长
Objective To observe the inhibitory effect of somatostatin on the growth of human pancreatic cancer cell line HS76 6T and to explore its mechanism. Methods The cells were cultured and added with different concentrations of somatostatin. The proliferation of cells was observed by MTT method. The content of intracellular cAMP was determined by radioimmunoassay. The concentration of intracellular Ca2 + was determined by Fura2 AM. Results Different concentrations of somatostatin (10 - 6 ~ 10 - 1 2 M) could effectively inhibit the growth of HS76 6T and inhibit the production of cAMP and Ca2 + in HS76 6T cells at 5 min and 3 min, respectively , cAMP production and Ca2 + levels and somatostatin concentration was negatively correlated. It enables the reduction of intracellular cAMP production and Ca2 + induced by pituitary adenylate cyclase activating polypeptide (PACAP). Conclusion Somatostatin can inhibit the growth of human pancreatic carcinoma HS76 6T cells and significantly inhibit the growth of PACAP-induced cells
