水稻簇矮病毒(RBSV)是由黑尾叶蝉(Nephotetixcincticeps)和二点黑尾叶蝉(N.virescens)传播的持久性病毒,但不经卵传递;病害表现有奇特的多蘖症或节枝症,且其病株汁液与水稻矮缩病毒(RDV)抗血清不起免疫反应.采用差速离心和蔗糖密度梯度离心,可获得RBSV提纯制剂,提纯制剂经紫外扫描,呈典型的核蛋白吸收曲线,A260/280=1.64.在电镜下观察提纯病毒,可见大量比较均一的球状粒体,直径为60.0nm.病毒粒体有双层衣壳,蛋白亚基清晰可辨.通过免疫扩散和免疫电镜试验,病毒粒体与RBSV抗血清有明显的血清学反应,而这种抗血清与RDV、水稻瘤矮病毒(RGDV)不起反应.提纯RBSV制剂以苯酚-甲基苯酚-SDS方法提取核酸.将其注射到昆虫介体内,具有侵染性.核酸制品有典型的核酸紫外吸收曲线,A260/280=2.02.根据核酸在不同离子强度下对RNase的稳定性等的测定,表明其核酸属双链RNA.根据紫外吸收测定,RNA在RBSV粒体中的含量为17.5%.用聚丙烯酰胺凝胶电泳测定,其RNA的总分子质量为16.66×106.各组分相对分子质量分别为2.70×106、2.30×106、1?
Rice cluster dwarf virus (RBSV) is a persistent virus that is transmitted by Nephotetix cincticeps and N. virescens but is not transmitted by eggs; the disease manifests as strange tillering or disease Strain, and its diseased juice and rice dwarf virus (RDV) antiserum not immune response. Using differential centrifugation and sucrose density gradient centrifugation, the RBSV purified preparation was obtained. The purified preparation was characterized by ultraviolet scanning and showed a typical nuclear protein absorption curve with A260 / 280 = 1.64.
