研究中介素(intermedin,IMD)基因在肾血管性高血压大鼠肥大心肌中的表达,以及缬沙坦、氨氯地平、依那普利三种不同药物降压后心肌中IMD基因表达的变化。用60只SD雄性大鼠随机分成假手术(作为对照组)和手术组,后者不完全性结扎左侧肾动脉,4周后又将手术组中成功建立肾性高血压模型的30只大鼠随机分为单纯结扎组和药物干预组,后者再分为三组,分别给于缬沙坦、氨氯地平、依那普利。药物干预10周后测定鼠尾动脉血压、心肌横径、左心室质量指数,RT-PCR半定量法检测心肌中IMD mRNA水平表达。结果显示单纯结扎组大鼠尾动脉收缩压较对照组明显升高,左室心肌横径、左室质量指数显著增加,左心室肌IMD mRNA表达也明显上调。缬沙坦、氨氯地平、依那普利干预组的血压、左室心肌横径、左室质量指数以及IMD mRNA表达显著低于单纯结扎组,但仍高于对照组,且三组间无显著差异。结论提示肾动脉不全结扎致高血压模型的肥大心肌中IMD mRNA得以上调,这可能改善左心室肥厚的病理生理过程,缬沙坦、氨氯地平和依那普利分别干预后,血压及肥大心肌改善,且心肌IMD mRNA表达均可以下调,提示心肌IMD mRNA的表达与血压或心肌肥大密切相关,未因降压药物的种类不同而表现出差异。
To study the expression of intermedial (IMD) gene in hypertrophic myocardium of renovascular hypertensive rats and the expression of IMD gene in myocardium after depressurization of three different drugs valsartan, amlodipine and enalapril Variety. The IMD mRNA expression in left ventricular muscle was also significantly increased. The valsartan, amlodipine, and enalapril intervention groups had significantly lower blood pressure, left ventricular myocardial diameter, left ventricular mass index and IMD mRNA expression than those in the simple ligation group, but still higher than those in the control group Significant differences. Conclusions The results suggest that IMD mRNA may be upregulated in hypertrophic myocardium induced by incomplete renal artery ligation, which may improve the pathophysiological process of left ventricular hypertrophy.
