目的:探讨金铁锁总皂苷(TSPT)镇痛作用及其机制,进一步揭示金铁锁总皂苷抗类风湿关节炎的药理作用基础。方法:采用热板法初步评价TSPT的镇痛效果,建立大鼠佐剂性关节炎(AA)模型,运用免疫组化研究TSPT对c-fos基因表达产物FOS蛋白水平的影响。结果:TSPT具有明显的镇痛作用,大剂量组优于中药阳性对照组;TSPT还能明显抑制AA大鼠炎性疼痛中枢及外周组织中c-fos基因的表达。结论:TSPT具有良好的镇痛效果,其镇痛作用可能与抑制中枢和外周神经c-fos基因的表达有关,为深入研究其抗类风湿关节炎作用奠定了基础。
Objective: To investigate the analgesic effect of TSPT and its mechanism, and to further reveal the pharmacological effects of the total saponins from Jinshuizhong against rheumatoid arthritis. Methods: The hot plate method was used to evaluate the analgesic effect of TSPT. The rat model of adjuvant arthritis (AA) was established. The effect of TSPT on the expression of Fos protein in c-fos gene was studied by immunohistochemistry. Results: TSPT had obvious analgesic effect. The high dose group was better than the traditional Chinese medicine positive control group. TSPT also could significantly inhibit the expression of c-fos gene in the central and peripheral tissues of inflammatory pain in AA rats. CONCLUSION: TSPT has a good analgesic effect. Its analgesic effect may be related to the inhibition of c-fos gene expression in central and peripheral nerves, which lays the foundation for further study on its anti-rheumatoid arthritis effect.
