目的:探讨PTEN在急性白血病中的表达情况及其与临床的关系。方法:用S-P免疫细胞化学方法检测52例初治急性白血病(AL),10例完全缓解期急性白血病(AL-CR)及14例正常对照(骨髓正常的非恶性血液病患者)骨髓单个核细胞中PTEN的表达并分析了PTEN与临床指标的相关性。结果:初治AL组PTEN的表达阳性率为46.2%,AL-CR组的表达阳性率为90%,正常对照组的表达阳性率为92.9%,初治组与AL-CR组,正常对照组比较差异有显著性(P<0.05),ANLL组表达阳性率为44.4%,ALL组表达阳性率为50%,ANLL组与ALL组比较差异无统计学意义(P>0.05),AL-CR组与正常对照组比较差异无统计学意义(P>0.05)。PTEN蛋白的低表达与患者的年龄、性别无明显相关(P>0.05)而与外周血白细胞计数及髓外浸润呈正相关(P<0.05)。结论:AL患者存在PTEN蛋白表达缺失,PTEN蛋白在AL的发病中可能起一定作用。
Objective: To investigate the expression of PTEN in acute leukemia and its clinical significance. Methods: 52 cases of newly diagnosed acute leukemia (AL), 10 complete remission acute leukemia (AL-CR) and 14 normal control (normal bone marrow non-hematologic malignancies) bone marrow mononuclear cells were detected by SP immunocytochemistry PTEN expression and analysis of PTEN and clinical indicators of relevance. Results: The positive expression rate of PTEN was 46.2% in primary AL group, 90% in AL-CR group and 92.9% in normal control group. Compared with AL-CR group, normal control group The positive rate was 44.4% in ANLL group and 50% in ALL group, but there was no significant difference between ANLL group and ALL group (P> 0.05). The low expression of PTEN protein had no significant correlation with age and sex of patients (P> 0.05), but positively correlated with peripheral blood leukocyte count and extramedullary infiltration (P <0.05). CONCLUSION: PTEN protein is absent in patients with AL, and PTEN protein may play a role in the pathogenesis of AL.
