目的 探讨不同剂量的抗氧化剂复合物 ,大豆异黄酮、维生素 C和维生素 E的协同作用对 2型糖尿病 (T2 DM)病人糖负荷时血管内皮炎性细胞因子表达的影响。方法 观察正常人对照组、糖尿病对照组和 3个不同剂量复合抗氧化剂 T2 DM病人干预组在口服葡萄糖耐量试验前、试验后 1 h、2 h、4h、6h血清中肿瘤坏死因子 α(TNF- α)和白细胞介素 6(IL- 6)的水平。结果 4个 T2 DM组空腹时 TNF- α和 IL- 6水平均高于正常对照组 ,其中 TNF-α水平四个 T2 DM组与正常对照组比较有显著性差异 (P<0 .0 1 )。口服葡萄糖耐量试验后 ,复合抗氧化剂呈剂量依赖性部分逆转T2 DM病人高血糖过氧化压力损伤下血管内皮细胞 TNF- α、IL- 6的表达。结论 不同剂量的天然抗氧化剂均可通过抑制 TNF- α和 IL- 6的表达来保护血管内皮细胞免受高血糖过氧化压力的过氧化损伤 ,从而达到防治或延缓动脉粥样硬化 (AS)的目的。其中 ,由 1 50 mg大豆异黄酮和 50 0 mg VC、40 0 IU VE组成的混合干预物效果最佳且剂量较低 ,可作为 T2 DM人群以防治等并发症为目的的适宜摄入量。
Objective To investigate the synergistic effects of different dosages of antioxidant complex, soy isoflavone, vitamin C and vitamin E on the expression of inflammatory cytokines in endothelium of type 2 diabetic patients undergoing glucose load. Methods The levels of tumor necrosis factor-α (TNF-α) in serum of normal control group, diabetic control group and 3 DM T2DM patients with different dosages of antioxidants before oral glucose tolerance test, 1 h, 2 h, 4 h, α) and interleukin 6 (IL-6) levels. Results The levels of TNF-α and IL-6 in the four T2 DM groups were all higher than those in the normal control group. There was a significant difference between the four T2 DM groups in TNF-α level and the normal control group (P <0.01) . After oral glucose tolerance test, the composite antioxidants partially reversed the dose-dependent partial reversal of TNF-α and IL-6 expression in vascular endothelial cells under hyperglycemic stress in T2DM patients. Conclusion Different dosages of natural antioxidants can protect vascular endothelial cells against peroxidative damage caused by hyperglycemic peroxidation by inhibiting the expression of TNF-α and IL-6, so as to prevent or delay atherosclerosis (AS) purpose. Among them, the mixed intervention consisting of 150 mg soy isoflavones and 50 0 mg VC and 40 0 IU VE had the best effect and low dosage, which could be regarded as suitable intake for prevention and treatment of T2DM population.
