目的为了研究肝细胞癌的发病机制,探讨细胞周期相关激酶(CCRK)在肝细胞癌组织中的表达及意义,以及CCRK与乙肝病毒X蛋白(HBx)之间的关系。方法 RT-PCR法和Western印迹法分别从mRNA水平和蛋白质水平检测CCRK和HBx在肝癌组织、癌旁组织和正常组织中的表达情况。结果肝癌组织中CCRK mRNA及蛋白水平均明显高于癌旁肝组织,差异具有统计学意义(P<0.05);CCRK的表达与肝癌患者的性别、年龄,HBsAg,肿瘤大小均无关,而与临床分期有关;CCRK与HBx没有相关性。结论 CCRK在肝癌组织中的表达水平高于癌旁组织,对肝细胞癌的发生发展具有促进作用,但是与HBx没有相关性。
Objective To investigate the pathogenesis of hepatocellular carcinoma (HCC), and to explore the expression and significance of CCRK in hepatocellular carcinoma (HCC) and the relationship between CCRK and hepatitis B virus X protein (HBx). Methods RT-PCR and Western blotting were used to detect the expression of CCRK and HBx in hepatocellular carcinoma, paracancerous tissues and normal tissues at mRNA and protein levels respectively. Results The expression of CCRK mRNA and protein in HCC tissues were significantly higher than that in adjacent non-cancerous liver tissues (P <0.05). The expression of CCRK was not related to the gender, age, HBsAg, tumor size, Staging related; CCRK and HBx no correlation. Conclusion The expression level of CCRK in HCC tissue is higher than that in adjacent tissues, which may promote the occurrence and development of HCC. However, it has no correlation with HBx.
