目的探讨砷中毒对哮喘患儿外周血分离T细胞凋亡的影响。方法选取38例哮喘患儿为观察组,38例健康儿童为对照组。采集两组的外周血,对T细胞进行分离,分别采用地塞米松或三氧化二砷(As2O3)进行24h的培养,观察T细胞凋亡情况。结果不加地塞米松和As2O3,观察组的细胞凋亡率明显低于对照组,差异有统计学意义(P<0.05);加入As2O3后,两组的细胞凋亡率与不加地塞米松和As2O3相比,均明显上升,差异有统计学意义(P<0.05);加入地塞米松后,两组的细胞凋亡率与不加地塞米松和As2O3相比,差异有统计学意义(P<0.05)。结论 As2O3可显著提高哮喘患儿外周血T细胞凋亡率,降低IL-4水平,减弱Bcl-2的表达。
Objective To investigate the effect of arsenic poisoning on the apoptosis of T cells isolated from peripheral blood of asthmatic children. Methods 38 children with asthma were selected as observation group and 38 healthy children as control group. The peripheral blood of the two groups were collected and T cells were isolated. The cells were cultured with dexamethasone or arsenic trioxide (As2O3) for 24h, and the apoptosis of T cells was observed. Results Without dexamethasone and As2O3, the apoptotic rate of the observation group was significantly lower than that of the control group, the difference was statistically significant (P <0.05); after adding As2O3, the apoptosis rate of the two groups was significantly higher than that of the non-dexamethasone and As2O3 (P <0.05). After adding dexamethasone, the apoptosis rates of the two groups were statistically significant compared with those without dexamethasone and As2O3 (P <0.05), the difference was statistically significant ). Conclusion As2O3 can significantly increase the rate of apoptosis of peripheral blood T lymphocytes in asthmatic children, decrease the level of IL-4 and decrease the expression of Bcl-2.
