目的:建立肝宁颗粒中川芎的鉴别与阿魏酸(FA)的含量测定方法。方法:采用TLC法鉴别川芎;采用HPLC法测定FA的含量。色谱柱:Kromasil ODS-1 C18(200mm×4.6mm,5μm);流动相:甲醇-水-冰醋酸(30∶69.5∶0.5);检测波长:322nm;柱温:室温;流速:1.0mL·min-1。结果:供试品TLC中,在与川芎对照药材及FA对照品色谱相应的位置上,显相同颜色斑点;FA进样量在0.01012~0.15180μg范围内与色谱峰峰面积积分值呈良好线性关系(r=0.9999,n=8);平均回收率为97.39%(RSD=1.97%)。结论:该鉴别与含量测定方法快速、准确、专属性强、重现性好,可用于肝宁颗粒的质量控制。
Objective: To establish the identification of Chuanxiong in Ganning Granule and the determination of ferulic acid (FA). Methods: The TLC method was used to identify Chuanxiong. The content of FA was determined by HPLC. Column: Kromasil ODS-1 C18 (200 mm×4.6 mm, 5 μm); mobile phase: methanol-water-glacial acetic acid (30:69.5:0.5); detection wavelength: 322 nm; column temperature: room temperature; flow rate: 1.0 mL·min -1. RESULTS: In the TLC of the test sample, the spots with the same color were found at the positions corresponding to the chromatograms of the Chuanxiong reference drug and the FA reference substance; the linear relationship between the injection volume of the FA in the range of 0.01012 to 0.15180 μg and the integrated value of the peak area of the chromatogram peak was good. (r=0.9999, n=8); The average recovery was 97.39% (RSD=1.97%). Conclusion: The method of identification and content determination is rapid, accurate, specific and reproducible, and can be used for quality control of Ganning Granules.
