M-sol保存液添加L-肉碱对血小板体外保存的影响

目的 探讨在M-sol保存液中添加L-肉碱(LC)对体外保存血小板的影响.方法 采集无偿献血志愿者的全血,采用白膜法制备浓缩血小板.将8袋ABO同型的浓缩血小板汇集,配制M-sol为血小板保存液.依据保存介质和LC 浓度不同分为6组,即血浆组、M-sol 组、M-sol+0.5mmol/L LC 组、M-sol+5mmol/L LC 组、M-sol+15mmol/L LC组和M-sol+25mmol/LLC组.置于(22±2)℃持续振荡保存,分别于保存d0、3、7取样.用全自动血细胞分析仪检测血小板计数(Plt)、平均血小板体积(MPV)、血小板分布宽度(PDW),用流式细胞仪检测血小板膜外磷脂酰丝氨酸(PS)和CD62p的阳性率,用半自动生化分析仪检测葡萄糖和乳酸含量,并计算保存7d期间葡萄糖消耗量和乳酸生成量.结果 随保存时间的延长,各组Plt均下降,MPV和PDW均升高,在同一时间点各组间差异无统计学意义(P>0.05).PS和CD62p阳性率逐渐升高,保存d7时,M-sol+15mmol/L LC组PS和CD62p阳性率低于其他各组(P<0.05).葡萄糖消耗量和乳酸生成量M-sol+15mmol/LLC组最低(P<0.05).结论 将合适浓度的LC加入M-sol血小板保存液中,可降低血小板贮存损伤.
